The earliest fortified sites in the kingdom of Judah in the early 10th century BCE feature a casemate city wall lined with an abutting belt of houses, which incorporate the casemates as rear rooms. This urban plan is clearly recognized in the sites of Khirbet Qeiyafa, Tell en-Nabeh, Khirbet ed-Dawwara, and, as discussed in detail, Beth Shemesh. Recently, excavations at Lachish, Level V, uncovered a similar pattern comprising a peripheral belt of structures abutting the city wall. This city wall was solid with no casemates. These sites have far-reaching implications for understanding the urbanization process, urban planning, and borders of the earliest phase of the kingdom of Judah.

SUMMARY Late blight caused by the oomycete Phytophthora infestans is a most devastating disease of potatoes (Solanum tuberosum). Its early detection is crucial for suppressing disease spread. Necrotic lesions are normally seen in leaves at 4 dpi (days post inoculation) when colonized cells are dead, but early detection of the initial biotrophic growth stage, when the pathogen feeds on living cells, is challenging. Here, the biotrophic growth phase of P. infestans was detected by whole-plant redox imaging of potato plants expressing chloroplast-targeted reduction-oxidation sensitive green fluorescent protein (chl-roGFP2). Clear spots on potato leaves with a lower chl-roGFP2 oxidation state were detected as early as 2 dpi, before any visual symptoms were recorded. These spots were particularly evident during light-to-dark transitions and reflected the mislocalization of chl-roGFP2 outside the chloroplasts. Image analysis based on machine learning enabled systematic identification and quantification of spots and unbiased classification of infected and uninfected leaves in inoculated plants. Comparing redox to chlorophyll fluorescence imaging showed that infected leaf areas which exhibit mislocalized chl-roGFP2 also showed reduced non-photochemical quenching (NPQ) and enhanced quantum PSII yield (ΦPSII) compared to the surrounding leaf areas. The data suggest that mislocalization of chloroplast-targeted proteins is an efficient marker of late blight infection and demonstrate how it can be utilized for nondestructive monitoring of the disease biotrophic stage using whole-plant redox imaging.

It is now well established that changes in the zonal wind stress over the ACC do not lead to changes in its baroclinicity nor baroclinic transport, a phenomenon referred to as “eddy saturation”. Previous studies provide contrasting dynamical mechanisms for this phenomenon: on one extreme, changes in the winds lead to changes the efficiency with which transient eddies transfer momentum to the sea floor; on the other, structural adjustments of the ACC’s standing meanders increase the efficiency of momentum transfer. In this study the authors investigate the relative importance of these mechanisms using an idealized, isopycnal channel model of the ACC. Via separate diagnoses of the model’s time-mean flow and eddy diffusivity, the authors decompose the model’s response to changes in wind stress into contributions from transient eddies and the mean flow. A key result is that holding the transient eddy diffusivity constant while varying the mean flow very closely compensates changes in the wind stress, whereas holding the mean flow constant and varying the eddy diffusivity does not. This implies that “eddy saturation” primarily occurs due to adjustments in the ACC’s standing waves/meanders, rather than due to adjustments of transient eddy behavior. The authors derive a quasi-geostrophic theory for ACC transport saturation by standing waves, in which the transient eddy diffusivity is held fixed, and thus provides dynamical insights into standing wave adjustment to wind changes. These findings imply that representing eddy saturation in global models requires adequate resolution of the ACC’s standing meanders, with wind-responsive parameterizations of the transient eddies being of secondary importance.

In the current review, we focused on identifying aliment compounds and micronutrients, as well as addressed promising bioactive nutrients that may interfere with NAFLD advance and ultimately affect this disease progress. In this regard, we targeted: 1. Potential bioactive nutrients that may interfere with NAFLD, specifically dark chocolate, cocoa butter, and peanut butter which may be involved in decreasing cholesterol concentrations. 2. The role of sweeteners used in coffee and other frequent beverages; in this sense, stevia has proven to be adequate for improving carbohydrate metabolism, liver steatosis, and liver fibrosis. 3. Additional compounds were shown to exert a beneficial action on NAFLD, namely glutathione, soy lecithin, silymarin, Aquamin, and cannabinoids which were shown to lower the serum concentration of triglycerides. 4. The effects of micronutrients, especially vitamins, on NAFLD. Even if most studies demonstrate the beneficial role of vitamins in this pathology, there are exceptions. 5. We provide information regarding the modulation of the activity of some enzymes related to NAFLD and their effect on this disease. We conclude that NAFLD can be prevented or improved by different factors through their involvement in the signaling, genetic, and biochemical pathways that underlie NAFLD. Therefore, exposing this vast knowledge to the public is particularly important.

Studies of electoral geography have traditionally examined the impact of spatial context on vote choice at the neighborhood or larger geographical level, overlooking potential effects of the immediate physical environment in the polling station. Observations of actual polling stations located in schools in Israel revealed a strong presence of nationalist and religious content in and around voting booths, in the form of naïve décor such as murals and children’s drawings (Study 1). In three experimental studies (Studies 2–4), we examine the effect on voters of such seemingly apolitical cues. The experimental studies were conducted in the days prior to Israeli general elections for the 21st and 22nd Knesset. Using a virtual-reality interface based on real-life content in actual polling stations (Study 2, student sample) and simulated environments (Studies 3 and 4, representative samples of Jewish Israeli voters), the three experiments document an effect of naïve nationalist décor on simulated voting, particularly for left-wing less-nationalist voters. In Study 5, based on actual voting in the 22nd Knesset, we capitalize on the random allocation of voters to polling stations in schools and find a correlation between the content displayed around polling places and voting patterns among distinctively left-wing populations. Investigating the influence of the encounter with the immediate environment at a resolution of meters and seconds exposes the potential impact of the (ultra-)micro temporo-spatial scale on decision-making and enriches theoretical discussions on the multiscalarity of contexts in electoral geography analyses.

The fusion step in the formation of colloidal quantum dot molecules, constructed from two core/shell quantum dots,Adar 2023 dictates the coupling strength and hence their properties and enriched functionalities compared to monomers. Herein, studying the monomer size effect on fusion and coupling, we observe a linear relation of the fusion temperature with the inverse nanocrystal radius. This trend, similar to that in nanocrystal melting, emphasizes the role of the surface energy. The suggested fusion mechanism involves intraparticle ripening where atoms diffuse to the reactive connecting neck region. Moreover, the effect of monomer size and neck filling on the degree of electronic coupling is studied by combined atomistic-pseudopotential calculations and optical measurements, uncovering strong coupling effects in small QD dimers, leading to significant optical changes. Understanding and controlling the fusion and hence coupling effect allows tailoring the optical properties of these nanoscale structures, with potential applications in photonic and quantum technologies.
Prolonged cannabis users show a lower prevalence of obesity and associated comorbidities. In rodent models, Δ9-tetrahydrocannabinol (THC) and cannabidiol (CBD) from the plant Cannabis sativa L. have shown anti-obesity properties, suggesting a link between the endocannabinoid system (ECS) and obesity. However, the oral administration route has rarely been studied in this context. The aim of this study was to investigate the effect of prolonged oral administration of pure THC and CBD on obesity-related parameters and peripheral endocannabinoids. C57BL/6 male mice were fed with either a high-fat or standard diet and then received oral treatment in ramping doses, namely 10 mg/kg of THC or CBD for 5 weeks followed by 30 mg/kg for an additional 5 weeks. Mice treated with THC had attenuated weight gain and improved glucose tolerance, followed by improvement in steatosis markers and decreased hypertrophic cells in adipose epididymal tissue. Mice treated with CBD had improved glucose tolerance and increased markers of lipid metabolism in adipose and liver tissues, but in contrast to THC, CBD had no effect on weight gain and steatosis markers. CBD exclusively decreased the level of the endocannabinoid 2-arachidonoylglycerol in the liver. These data suggest that the prolonged oral consumption of THC, but not of CBD, ameliorates diet-induced obesity and metabolic parameters, possibly through a mechanism of adipose tissue adaptation.

The current study analyzes the cause of the extreme change in dermal permeation resulting from a minute structural transformation within a temperature-sensitive direct hexagonal mesophase by a multiple-method approach. The evaluated mesophase is composed of Tween 80, propylene glycol, isopropyl myristate, benzyl alcohol, and water. 1 wt% ketoconazole (KCZ) was loaded in the concentrate as a model lipophilic drug. Evaluation of the mesophase at 25 °C indicated that the hexagonal mesophase region (33.0–42.5 wt% water along water dilution line N91) consists of two distinct structures. At water content below 39.0 wt%, the mesophase is structured as a bidiscontinuous hexagonal mesophase (BHM) consisting of occluded hydration centers and hexagonally oriented direct micelles. Above 39.5 wt% water, dilution of the BHM obstructs the hydration centers, transforming the mesophase to a water continuous hexagonal mesophase (CHM). FTIR evaluation indicated that the both the added water upon dilution and the water molecules released from the hydration centers bind to the surfactant's non-hydrated ethoxylates. Evaluation of the structural transition by PGSE-NMR and DSC indicated that the binding of water to the surfactant's heads immobilizes the inner core of the solubilized lipids. The reduction of the lipid core’s mobility prevents its crystallization at cryogenic temperatures. SAXS and rheological evaluations pointed out that both BHM and CHM are temperature-sensitive mesophases, which lose their range order and storage modulus at 32 °C. Obstruction of the hexagonal mesophase enhances KCZ's diffusion from both mesophases’ via cellulose acetate at 35 °C. Despite the similarity of both mesophases at elevated temperatures (35 °C), the lipidic inner core’s rotational correlation time for deformed CHM micelles remains substantially lower than from the deformed BHM. Franz cell evaluation indicated that the lipidic core mobility substantially influences KCZ's distribution upon dermal application, whereas from CHM KCZ's transdermal flux via a dermatome bovine graft at 35 °C, was 1.60 ± 0.28 µg/cm2 h, no flux was detected from BHM. On the other hand, KCZ's dermal accumulation from BHM, at 35 °C, was found to be more than double than that from CHM, indicating that the decrease in the lipidic inner core’s mobility of direct micelles prevents the accumulation of lipophilic compounds in the dermis, thereby enhancing their transdermal flux.

In mediated interactions (e.g. video calls), less information is available about the other. To investigate how this affects our empathy for one another, we conducted an electroencephalogram study, in which 30 human participants observed 1 of 5 targets undergoing painful electric stimulation, once in a direct interaction and once in a live, video-mediated interaction. We found that observers were as accurate in judging others' pain and showed as much affective empathy via video as in a direct encounter. While mu suppression, a common neural marker of empathy, was not sensitive to others' pain, theta responses to others' pain as well as skin conductance coupling between participants were reduced in the video-mediated condition. We conclude that physical proximity with its rich social cues is important for nuanced physiological resonance with the other's experience. More studies are warranted to confirm these results and to understand their behavioral significance for remote social interactions

Background: Viral infection is associated with a significant rewire of the host metabolic pathways, presenting attractive metabolic targets for intervention. Methods: We chart the metabolic response of lung epithelial cells to SARS-CoV-2 infection in primary cultures and COVID-19 patient samples and perform in vitro metabolism-focused drug screen on primary lung epithelial cells infected with different strains of the virus. We perform observational analysis of Israeli patients hospitalized due to COVID-19 and comparative epidemiological analysis from cohorts in Italy and the Veteran's Health Administration in the United States. In addition, we perform a prospective non-randomized interventional open-label study in which 15 patients hospitalized with severe COVID-19 were given 145 mg/day of nanocrystallized fenofibrate added to the standard of care. Results: SARS-CoV-2 infection produced transcriptional changes associated with increased glycolysis and lipid accumulation. Metabolism-focused drug screen showed that fenofibrate reversed lipid accumulation and blocked SARS-CoV-2 replication through a PPARα-dependent mechanism in both alpha and delta variants. Analysis of 3233 Israeli patients hospitalized due to COVID-19 supported in vitro findings. Patients taking fibrates showed significantly lower markers of immunoinflammation and faster recovery. Additional corroboration was received by comparative epidemiological analysis from cohorts in Europe and the United States. A subsequent prospective non-randomized interventional open-label study was carried out on 15 patients hospitalized with severe COVID-19. The patients were treated with 145 mg/day of nanocrystallized fenofibrate in addition to standard-of-care. Patients receiving fenofibrate demonstrated a rapid reduction in inflammation and a significantly faster recovery compared to patients admitted during the same period. Conclusions: Taken together, our data suggest that pharmacological modulation of PPARα should be strongly considered as a potential therapeutic approach for SARS-CoV-2 infection and emphasizes the need to complete the study of fenofibrate in large randomized controlled clinical trials.
