Most of what we currently know about how neural circuits work we owe to methods based on the electrical or optical recording of neural activity. This is changing dramatically. First, the advent of optogenetic techinques has enabled precise manipulation of the activity of specific neurons. Second, the development of super-resolution methods for obtaining detailed maps of synaptic connectivity has paved the way for uncovering the connectomes of entire brains or brain regions. We describe a third and complementary new strategy for investigating and manipulating neural circuits: the artificial insertion of new synapses into existing neural circuits using genetic engineering tools. We have successfully accomplished this in C. elegans. Thus, In addition to being the first animal with an entirely mapped connectome, C. elegans is now also the first animal to have an editable connectome. Variations on this approach may be applicable in more complex nervous systems

תקציר The purpose of this work was to investigate whether low-frequency, low-intensity (20 kHz, <100 mW/cm(2), spatial-peak, temporal-peak intensity) ultrasound, delivered with a lightweight (<100 g), tether-free, fully wearable, battery-powered applicator, is capable of reducing inflammation in a mouse model of rheumatoid arthritis. The therapeutic, acute, anti-inflammatory effect was estimated from the relative swelling induced in mice hindlimb paws. In an independent, indirect approach, the inflammation was bio-imaged by measuring glycolytic activity with near-infrared labeled 2-deoxyglucose. The outcome of the experiments indicated that the combination of ultrasound exposure and topical application of 0.1% (w/w) betamethasone gel resulted in statistically significantly (p < 0.05) enhanced anti-inflammatory activity in comparison with drug or ultrasound treatment alone. The present study underscores the potential benefits of low-frequency, low-intensity ultrasound-assisted drug delivery. However, the proof of concept presented indicates the need for additional experiments to systematically evaluate and optimize the potential of, and the conditions for, tolerable low-frequency, low-intensity ultrasound-promoted non-invasive drug delivery.

תקציר Permethrin, a poorly water-soluble synthetic pesticide belonging to the pyrethroid family, was formulated into water-dispersive nanometric form by rapid evaporation of pesticide loaded oil-in-water microemulsion. The mean hydrodynamic diameter of Nanopermethrin was found to be 199.01 ± 1.4 nm. The efficacy of the Nanopermethrin was comparatively investigated with its bulk form against 2-3 days old adult mosquitoes by WHO cone bioassay for 60 min. The median knockdown concentration of Culex tritaeniorhynchus, Culex quinquefasciatus and Aedes albopictus were found to be 7.20 × 10(4), 7.53 × 10(4), 0.42 × 10(3) mg/L for Bulk permethrin, and 0.98 × 10(4), 1.17 × 10(4), 0.05 × 10(3) mg/L for Nanopermethrin, respectively. The obtained results extrapolate the improved efficacy of Nanopermethrin even at low-level concentrations. Hence, the formulated Nanopermethrin will serve as an effective alternative pesticide in controlling the mosquito population with reduced environmental toxicity.[on SciFinder (R)]

Quantum heat engines (QHE) are thermal machines where the working substance is a quantum object. In the extreme case, the working medium can be a single particle or a few-level quantum system. The study of QHE has shown a remarkable similarity with macroscopic thermodynamical results, thus raising the issue of what is quantum in quantum thermodynamics. Our main result is the thermodynamical equivalence of all engine types in the quantum regime of small action with respect to Planck’s constant. They have the same power, the same heat, and the same efficiency, and they even have the same relaxation rates and relaxation modes. Furthermore, it is shown that QHE have quantum-thermodynamic signature; i.e., thermodynamic measurements can confirm the presence of quantum effects in the device. We identify generic coherent and stochastic work extraction mechanisms and show that coherence enables power outputs that greatly exceed the power of stochastic (dephased) engines.
In angiosperms, the production of flowers marks the beginning of the reproductive phase. At the emergence of flower primordia on the flanks of the inflorescence meristem, the WUSCHEL (WUS) gene, which encodes a homeodomain transcription factor starts to be expressed and establishes de novo stem cell population, founder of the floral meristem (FM). Similarly to the shoot apical meristem a precise spatial and temporal expression pattern of WUS is required and maintained through strict regulation by multiple regulatory inputs to maintain stem cell homeostasis. However, following the formation of a genetically determined fixed number of floral organs, this homeostasis is shifted towards organogenesis and the FM is terminated. In here we performed a genetic study to test how a reduction in ERECTA, CLAVATA and class III HD-ZIP pathways affects floral meristem activity and flower development. We revealed strong synergistic phenotypes of extra flower number, supernumerary whorls, total loss of determinacy and extreme enlargement of the meristem as compared to any double mutant combination indicating that the three pathways, CLV3, ER and HD-ZIPIII distinctively regulate meristem activity and that they act in parallel. Our findings yield several new insights into stem cell-driven development. We demonstrate the crucial requirement for coupling floral meristem termination with carpel formation to ensure successful reproduction in plants. We also show how regulation of meristem size and alternation in spatial structure of the meristem serve as a mechanism to determine flower organogenesis. We propose that the loss of FM determinacy due to the reduction in CLV3, ER and HD-ZIPIII activity is genetically separable from the AGAMOUS core mechanism of meristem termination.
