The p53 tumor suppressor protein is the most well studied as a regulator of transcription in the nucleus, where it exists primarily as a tetramer. However, there are other oligomeric states of p53 that are relevant to its regulation and activities. In unstressed cells, p53 is normally held in check by MDM2 that targets p53 for transcriptional repression, proteasomal degradation, and cytoplasmic localization. Here we discovered a hydrophobic region within the MDM2 N-terminal domain that binds exclusively to the dimeric form of the p53 C-terminal domain in vitro. In cell-based assays, MDM2 exhibits superior binding to, hyperdegradation of, and increased nuclear exclusion of dimeric p53 when compared with tetrameric wild-type p53. Correspondingly, impairing the hydrophobicity of the newly identified N-terminal MDM2 region leads to p53 stabilization. Interestingly, we found that dimeric mutant p53 is partially unfolded and is a target for ubiquitin-independent degradation by the 20S proteasome. Finally, forcing certain tumor-derived mutant forms of p53 into dimer configuration results in hyperdegradation of mutant p53 and inhibition of p53-mediated cancer cell migration. Gaining insight into different oligomeric forms of p53 may provide novel approaches to cancer therapy.

How do women travelling the colonial frontier create a feminine, and potentially less hierarchical type of modernity? And how does Neo-Victorian fiction explore gendered and racialized types of modernity through the use of travel? Eleanor Catton’s The Luminaries (2013) represesnts the quest for a postcolonial and feminine modernity through the trope of the woman traveler, worker, storyteller and entrepreneur. In particular, protagonists Anna Whetherell and Lydia Wells oppose the highly racist and sexist societies of gold rush frontier towns of the 1860s New Zealand through solitary travel on foot, by sea and across textual layers. This paper argues that such independent solitary women travelers stand for a new representation of white women in colonial contexts and challenge traditional categories of Victorian femininity, such as the dichotomous opposition between the Angel in the House and the fallen woman. By shifting across white femininity and queer Chinese identities (in Anna’s case), and by embracing a masculine, capitalist model (for Lydia), Catton’s heroines survive, on their own, as members of a minority in the communities of white, male miners. The two women thus embody new types of femininity and, while placing themselves outside the colonial hierarchy, they question the social structure, the exploitation of the Other (the woman, the Chinese) and set an example for a more viable and more equal society born out of colonial settlement. Finally, while shaping modernity through their female gaze and a free way of travelling the peripheries, the two women also accomplish their own Bildung process and, forgers of their own fortunes, symbolize the shift from masculine, imperial modernity to a feminine, neo-Victorian, postcolonial paradigm.

BACKGROUND: Current efforts to develop stem cell therapy as a novel treatment for neurointestinal diseases are limited by the unavailability of a model system to study cell transplantation in the human intestine. We propose that xenograft models support enteric nervous system (ENS) development in the fetal human intestine when transplanted into mice subcutaneously or intra-abdominally. METHODS: Fetal human small and large intestine were grafted onto the small intestinal mesentery and into the subcutaneous tissue of immunodeficient mice for up to 4 months. Intestinal cytoarchitecture and ENS development were studied using immunohistochemistry. KEY RESULTS: In both abdominal and subcutaneous grafts, the intestine developed normally with formation of mature epithelial and mesenchymal layers. The ENS was patterned in two ganglionated plexuses containing enteric neurons and glia, including cholinergic and nitrergic neuronal subtypes. c-Kit-immunoreactive interstitial cells of Cajal were present in the gut wall. CONCLUSIONS & INFERENCES: Abdominal xenografts represent a novel model that supports the growth and development of fetal human intestine. This in vivo approach will be a useful method to study maturation of the ENS, the pathophysiology of neurointestinal diseases, and the long-term survival and functional differentiation of neuronal stem cells for the treatment of enteric neuropathies.
