Cromolyn Sodium Differentially Regulates Human Mast Cell And Mouse Leukocyte Responses To Control Allergic Inflammation.. Pharmacological Research, Pp. 106172. doi:10.1016/j.phrs.2022.106172. תקציר
BACKGROUND: Cromolyn Sodium (CS) has been used in the past as an anti-allergy drug owing to its mast cell (MC) stabilizing properties that impair histamine release. However, additional mechanisms for its clinical actions are likely and might help to identify new roles for MCs and leukocytes in regulating inflammation. Here, using human cord blood-derived MCs (CBMCs), murine bone marrow-derived MCs (BMMCs) and eosinophils (BMEos), and in vivo mouse models of allergic inflammation (AI), additional actions of CS on MCs were determined. METHODS: The in vitro effects of CS on IgE-activated human and mouse MCs were assessed by measuring the levels of pro-inflammatory (tryptase, LTC(4), IL-8, CD48) and pro-resolution effectors (IL-10, CD300a, Annexin A1) before and after CS treatment. The in vivo effects of daily CS injections on parameters of inflammation were assessed using mouse models of allergic peritonitis (AP) (Ovalbumin/Alum- or Ovalbumin/S. aureus enterotoxin B) and allergic airways inflammation (AAI) (house dust mite (HDM)). RESULTS: In vitro, CS did not affect pro-inflammatory effectors but significantly increased the anti-inflammatory/pro-resolution CD300a levels and IL-10 release from IgE-activated CBMCs. BMMCs were not affected by CS. In vivo, CS injections decreased total cell and Eos numbers in the peritoneal cavity in the AP models and bronchoalveolar lavage and lungs in the AAI model. CS reduced EPX release from PAF-activated BMEos in vitro, possibly explaining the in vivo findings. CONCLUSION: Together, these results demonstrate immunomodulatory actions for CS in AI that are broader than only MC stabilization.
Cross-section measurements for a $Z$ boson produced in association with high-transverse-momentum jets ($p_\\mathrm\T\\ \geq 100$ GeV) and decaying into a charged-lepton pair ($e^+e^-,μ^+μ^-$) are presented. The measurements are performed using proton-proton collisions at $\sqrt\s\=13$ TeV corresponding to an integrated luminosity of $139$ fb$^\-1\$ collected by the ATLAS experiment at the LHC. Measurements of angular correlations between the $Z$ boson and the closest jet are performed in events with at least one jet with $p_\\mathrm\T\\ \geq 500$ GeV. Event topologies of particular interest are the collinear emission of a $Z$ boson in dijet events and a boosted $Z$ boson recoiling against a jet. Fiducial cross sections are compared with state-of-the-art theoretical predictions. The data are found to agree with next-to-next-to-leading-order predictions by NNLOjet and with the next-to-leading-order multi-leg generators MadGraph5_aMC@NLO and Sherpa.

Fruit skin reticulation is accompanied by the formation of a wound-periderm tissue made of suberized cells. The regulatory networks overseeing skin reticulation during fruit development were extensively studied, yet how reticulation affects post-harvest traits remains unknown. We addressed this notion using the common Cucumis sativus and the skin-cracked Sikkim (Cucumis sativus var. sikkimensis) cucumbers. Light and electron microscopy in consort with gas chromatography-mass spectrometry revealed that sativus fruit skin is made of the typical cutin polymer, while the skin of sikkimensis fruit comprised of the aromatic suberin polymer. Comparative post-harvest experiments with different storage temperatures revealed that sikkimensis fruit are more resilient to chilling injuries arise during cold storage, exhibiting lower rates of weight losses, ethylene and CO2, electrolyte leakage and lipid peroxidation. We further demonstrate that different storage temperatures affect the contents of skin polymers cutin and suberin in a differential manner.

PurposeThis study is to compare the impact of organizational agility and flexibility on performance of each type of product innovation (radical vs incremental innovation). Additionally, the moderating effect of technological turbulence on the relationship between the two types of organization is examined.Design/methodology/approachBased on gaps in the existing literature, the survey data are collected from managers who are in charge of developing new products in three industries: food and beverage, chemical and machinery (N = 431). Confirmatory factory analysis is used to verify measurement items and regression analysis is used to test hypotheses.FindingsThe results show that organizational agility increases performance in radical innovation both in a certain situation and an environment with technological turbulence. In contrast, the impact of organizational flexibility is limited to increasing performance in both radical and incremental innovation performance in a certain situation.Originality/valueOur study extends the knowledge of organizational agility and flexibility in the domain of product innovation. Adaptation of organization to respond the technological turbulence will stimulate creativity of new product development teams to produce new useful ideas and transform these ideas to product innovation. The different types of organizing a new product development team to handle technological turbulence will provide different results in product innovation performance. In addition, the findings provide a recommendation on how the organization of a new product development team can improve performance in each type of product innovation under technological turbulence.
This article engages three deities, one Greek and two Oriental, that their cults were worshipped in caves during the Hellenistic and Roman periods. The first deity is a Hellenistic terracotta figurine of Aphrodite, recovered from the prehistoric cave Me‘arat ha-Nahal (Wadi el-Maghara) at the foot of Mount Carmel. It probably represents Aphrodite Pandemos (Ἀϕροδίτη Πάνδημος) or Aphrodite en Kepois (Ἀϕροδίτη ἐν Κήποις). It may be assumed that the cave, and its proximity to the city of Dor, was modified to serve as a cultic site or shrine. The second deity is represented by a sunken relief engraved on a rough rock surface adjacent to a cluster of 18 caves, known as “The Temple Cave” complex, along Keziv Stream (Nahal Keziv) in western Galilee. The largest and main cave in this complex seems to have had a cultic function in the Roman period, that is, it constituted a cultic site for a particular divinity. The sunken relief depicts a walking male military figure, dubbed “The Man in the Wall.” Based on a comparative study and the figure’s iconographic characteristics, we may identify it with Sol Invictus Mithras, a Late Roman-period deity, manifesting cultic pagan activity in a remote and isolated area, in the very heart of nature. The third deity is Ba‘al Carmel (identified with Zeus/Jupiter) who was presumably worshipped in Elijah’s Cave on the western slope of Mt. Carmel. Ba‘al Carmel’s visual representation, the depiction of a libation vessel and the presumed figure of a priest or, alternatively, an altar within an aedicula suggest it was used in the Roman period. Notably, one of the Greek inscriptions, dated to the Roman period, explicitly addresses the cave’s sacred nature and the prohibition against its profanation.

An adverse drug reaction is defined as any noxious, unintended, and undesired effect of a drug that occurs at doses used in humans for prophylaxis, diagnosis, or therapy. A cutaneous eruption is one of the most common manifestations of an adverse drug reaction. This chapter reviews the epidemiology, etiology, diagnosis, clinical manifestations, and differential diagnosis of adverse drug reactions, as well as laboratory tests for them. Also discussed are the types of cutaneous eruption: exanthematous eruption, urticarial eruption, blistering eruption, pustular eruption, and others. The simple and complex forms of each type of eruption are reviewed. The chapter includes 4 tables and 12 figures. Tables present the warning signs of a serious drug eruption, clinical features of hypersensitivity syndrome reaction and serum sickness-like reaction, characteristics of Stevens-Johnson Syndrome and toxic epidermal necrolysis, and clinical pearls to identify anticoagulant-induced skin necrosis. Figures illustrate hypersensitivity syndrome reaction, a fixed drug eruption from tetracycline, pseudoporphyria from naproxen, linear immunoglobulin A disease induced by vancomycin, pemphigus foliaceus from taking enalapril, pemphigus vulgaris from taking penicillamine, toxic epidermal necrolysis after starting phenytoin therapy, acneiform drug eruption due to gefitinib, acute generalized exanthematous pustulosis from cloxacillin, coumarin-induced skin necrosis, a lichenoid drug eruption associated with ramipril, and leukocytoclastic vasculitis from hydrochlorothiazide. This chapter contains 106 references.

The COVID-19 outbreak is emerging as a significant public health challenge. Excessive production of proinflammatory cytokines, also known as cytokine storm, is a severe clinical syndrome known to develop as a complication of infectious or inflammatory diseases. Clinical evidence suggests that the occurrence of cytokine storm in severe acute respiratory syndrome secondary to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection is closely associated with the rapid deterioration and high mortality of severe cases. In this review, we aim to summarize the mechanism of SARS-CoV-2 infection and the subsequent immunological events related to excessive cytokine production and inflammatory responses associated with ACE2-AngII signaling. An overview of the diagnosis and an update on current therapeutic regimens and vaccinations is also provided.

Drawing on feminist, enactivist and posthumanist theories of reading, the essay develops theoretical and methodological tools for bodily and reflective reading of fictional figures. It introduces the notion of “readerly choreography,” which stands for the iterative experiential patterns that fictional narratives suggest. The primary purpose of the notion is to provide a better grasp of readerly dynamics typical to genre-derived works of fiction — including the cases in which generic frames of expectation and experience are estranged and reconfigured. The essay’s contribution to theory is presented on the basis of a reading of Paolo Bacigalupi’s “The People of Sand and Slag” (2004). This short story plays on the conventions of action-adventure, exaggerating the toughness and physical capabilities of technologically enhanced, posthuman action heroes. Owing to this exaggeration, it becomes difficult for readers to continue to perform the habitual experiential patterns of excitement, action-derived pleasure, and identification with the heroic protagonist. In other words, “The People of Sand and Slag” estranges the readerly choreography of action-adventure narratives.

D-Dimer Levels In Patients With Nonvalvular Atrial Fibrillation And Acute Heart Failure Treated With Edoxaban. J. Cardiol., 79, 6, Pp. 759–767. תקציר
BACKGROUND: D-dimer levels can predict ischemic stroke in patients with acute heart failure (AHF). However, the effects of direct oral anticoagulants on D-dimer levels have not been investigated during admission for AHF in patients with atrial fibrillation (AF). This study examined D-dimer levels immediately after admission and following edoxaban initiation as a sub-analysis of a multi-center study that investigated the pharmacokinetics and pharmacodynamics of edoxaban in patients with nonvalvular AF (NVAF) and AHF. METHODS: Hospitalized patients with NVAF and AHF received edoxaban according to the label. The primary measure was the change in D-dimer levels on 7 consecutive days after admission for AHF. We also investigated differences according to prior edoxaban use (de novo at the time of admission or continuation). RESULTS: In 10/13 (76.9%) de novo patients, D-dimer levels exceeded the reference value (1.0 µg/mL) at admission (mean, 2.12 µg/mL) and subsequently decreased in 9 patients (at final blood sampling: mean, 1.12 µg/mL); 1 patient did not fall below the reference value due to stasis dermatitis. In the continuation group, most patients had D-dimer levels below the reference value from Day 1 (mean, 0.93 µg/mL), and levels remained stable or decreased (at final blood sampling: mean, 0.49 µg/mL). No events of stroke were observed. CONCLUSIONS: D-dimer levels may be elevated in patients with NVAF and AHF, particularly in those without prior anticoagulant treatment. Edoxaban may be effective for lowering and keeping D-dimer levels, a biomarker for predicting ischemic stroke, below the reference value in patients with NVAF and AHF.
