Perinatal exposure to $Δ$(9)-tetrahydrocannabinol (THC) affects brain development and might increase the incidence of psychopathology later in life, which seems to be related to a dysregulation of endocannabinoid and/or dopaminergic systems. We here evaluated the transcriptional regulation of the genes encoding for the cannabinoid CB1 receptor (Cnr1) and the dopamine D2 receptor (Drd2) in perinatal THC-(pTHC) exposed male rats, focusing on the role of DNA methylation analyzed by pyrosequencing. Simultaneously, the molecular and behavioral abnormalities at two different time points (i.e., neonatal age and adulthood) and the potential preventive effect of peripubertal treatment with cannabidiol, a non-euphoric component of Cannabis, were assessed. The DRD2 methylation was also evaluated in a cohort of subjects with schizophrenia. We observed an increase in both Cnr1 and Drd2 mRNA levels selectively in the prefrontal cortex of adult pTHC-exposed rats with a consistent reduction in DNA methylation at the Drd2 regulatory region, paralleled by social withdrawal and cognitive impairment which were reversed by cannabidiol treatment. These adult abnormalities were preceded at neonatal age by delayed appearance of neonatal reflexes, higher Drd2 mRNA and lower 2-arachidonoylglycerol (2-AG) brain levels, which persisted till adulthood. Alterations of the epigenetic mark for DRD2 were also found in subjects with schizophrenia. Overall, reported data add further evidence to the dopamine-cannabinoid interaction in terms of DRD2 and CNR1 dysregulation which could be implicated in the pathogenesis of schizophrenia spectrum disorders, suggesting that cannabidiol treatment may normalize pTHC-induced psychopathology by modulating the altered dopaminergic activity.

Halide perovskite semiconductors are poised to revitalize the field of ionizing radiation detection as they have done to solar photovoltaics. We show that all-inorganic perovskite CsPbBr3 devices resolve 137Cs 662-keV γ-rays with 1.4% energy resolution, as well as other X- and γ-rays with energies ranging from tens of keV to over 1 MeV in ambipolar sensing and unipolar hole-only sensing modes with crystal volumes of 6.65 mm3 and 297 mm3, respectively. We report the scale-up of CsPbBr3 ingots to up to 1.5 inches in diameter with an excellent hole mobility–lifetime product of 8 × 10−3 cm2 V−1 and a long hole lifetime of up to 296 μs. CsPbBr3 detectors demonstrate a wide temperature region from ~2 °C to ~70 °C for stable operation. Detectors protected with suitable encapsulants show a uniform response for over 18 months. Consequently, we identify perovskite CsPbBr3 semiconductor as an exceptional candidate for new-generation high-energy γ-ray detection.

Eyal Ben-Dor Cohen, Ilan, Micha , ו Yarden, Oded . 2021. The Culturable Mycobiome Of Mesophotic Agelas Oroides: Constituents And Changes Following Sponge Transplantation To Shallow Water. Journal Of Fungi, 7, 7. doi:10.3390/jof7070567. תקציר
Marine sponges harbor a diverse array of microorganisms and the composition of the microbial community has been suggested to be linked to holo-biont health. Most of the attention concerning sponge mycobiomes has been given to sponges present in shallow depths. Here, we describe the presence of 146 culturable mycobiome taxa isolated from mesophotic niche (100 m depth)-inhabiting samples of Agelas oroides, in the Mediterranean Sea. We identify some potential in vitro interactions between several A. oroides-associated fungi and show that sponge meso-hyl extract, but not its predominantly collagen-rich part, is sufficient to support hyphal growth. We demonstrate that changes in the diversity of culturable mycobiome constituents occur following sponge transplantation from its original mesophotic habitat to shallow (10 m) waters, where historically (60 years ago) this species was found. We conclude that among the 30 fungal genera identified as associated with A. oroides, Aspergillus, Penicillium and Trichoderma constitute the core mycobiome of A. oroides, and that they persist even when the sponge is transplanted to a suboptimal environment, indicative of the presence of constant, as well as dynamic, components of the sponge mycobiome. Other genera seemed more depth-related and appeared or disappeared upon host's transfer from 100 to 10 m.

Cyclopentane modified FIT-PNA (cpFIT-PNA) probes are reported as highly emissive RNA sensors with the highest reported brightness for FIT-PNAs. Compared to FIT-PNAs, cpFIT-PNAs have improved mismatch discrimination for several pyrimidine-pyrimidine single nucleotide variants (SNVs).

In Heshels Kingdom, Dan Jacobson explores the impact of the British Empire’s expansion on Lithuanian Jewry. His memoir constructs a “mattering map” of the experience of his family, after the death of his grandfather, Heshel. Like more than thirty thousand other Jews, the bereaved family moved to a welcoming South Africa.

Heshels Kingdom is a split/screen account, alternating between Kimberley, South Africa, and Varniai, Lithuania. Their juxtaposition leads Dan Jacobson to chart the experiences of two Jewish communities, and construct a narrative map of familial and communal life. This split/screen account is not symmetrical. For the South Africa narrative, the narrator relies on familial and personal history. But for Lithuania he must tease out information from absence, seeking bits and remnants of the murdered Lithuanian Jewish community in order to find a purchase on which to reconstruct life in his grandfather’s Varniai, a small, Nazi-destroyed Lithuanian town.

The narrator interrogates the images of the two communities: Jacobson addresses the jacket-cover photograph of grandfather Heshel as if it might speak to him, and thus help him discover details of the life of his Lithuanian grandfather, whom he never knew. Asking questions, Jacobson invites the reader to engage with him as if they were looking together at a family-album: familial-networks begin to emerge, and kinship relationships elaborate the family’s life in South Africa; once activated the narrator can tease it into continuing the search for family experience. But the questions about Lithuania do not elicit much in the way of answers, for that Jewish community was destroyed by the Nazis and their Lithuanian helpers. Following the narrator’s lead, the reader’s imagination works to construct a comparative account both of the Jewish immigration to South Africa and the Jewish catastrophe in Lithuania, defining a “mattering map” of modern Jewish experience.

Researchers are increasingly using online audiences to conduct studies and surveys, but there is still considerable uncertainty about the quality of the available audience options. We first engage with the research community to explore which attributes of data quality are most critical to researchers: comprehension, attention, honesty, and reliability. We then explore differences in these data quality aspects between online platforms (where participants choose tasks) and online panels (where respondents are assigned surveys). We find that most audiences suffer concerning deficits, especially in comprehension and attention, except for Prolific, which appears superior to the other options on these and other parameters. The disappointing results from the other platforms and panels should alarm researchers who may be aggregating over serious noise in the quality of their results. Additionally, we provide a framework for ongoing investigation into the ever-changing nature of what aspects of data quality are important to researchers, and how the evolving set of audiences performs on these key aspects.
