This article deals with an obscure Biblical Hebrew (BH) wine-associated lexeme, ẓo'e ( צעה ). The lexeme appears five times as metaphors in the biblical Books of Isaiah and Jeremiah, reflecting consistency in the contemporary Judahite vernacular. The translations of these occurrences vary greatly, suggesting that the idiom may have fallen out of use after the Babylonian conquest in 586 BCE. The lexeme also appears on Arad Ostracon 16 (AO 16), which derives from the late Iron Age fortress of the same name. It was recently discovered thanks to the application of advanced technologies to the ostracon’s verso, revealing the lexeme הצע, hẓ'. In this article, we endorse the newly deciphered reading of AO 16 but reinterpret its content, offering new insight into wine handling in late Iron Age Judah.

This study sheds light on the central role of adenine nucleotide translocase 2 (ANT2) in the pathogenesis of obesity-induced CKD. Our data demonstrate that ANT2 depletion in renal proximal tubule cells (RPTCs) leads to a shift in their primary metabolic program from fatty acid oxidation to aerobic glycolysis, resulting in mitochondrial protection, cellular survival, and preservation of renal function. These findings provide new insights into the underlying mechanisms of obesity-induced CKD and have the potential to be translated toward the development of targeted therapeutic strategies for this debilitating condition.

SummaryMemory consolidation following learning is a dynamic and complex process comprising several transitions between distinct memory phases. Although memory consolidation has been studied extensively, it remains difficult to draw an integral description that can delimit the transition points between specific memory phases at the behavioral, neuronal, and genetic levels. To this end, we have developed a rapid and robust aversive conditioning protocol for the nematode worm Caenorhabditis elegans, tracing memory consolidation within the first hour post conditioning and then up to 18 h post conditioning. This made it possible to uncover time-dependent involvement of primary sensory neurons, transcription and translation processes, and diverse gene populations in memory consolidation. The change in neuronal valence was strong enough to induce second order conditioning, and was amenable to considerable modulation in specific mutant strains. Together, our work lends memory consolidation to detailed temporal and spatial analysis, advancing system-wide understanding of learning and memory.

Fever in infants presenting to pediatric emergency departments (PEDs) often results in significant return visits (RVs). This retrospective study aimed to identify factors associated with RVs in febrile infants aged 0 to 90 days. Data from infants presenting to PED between 2018 and 2021 and returning within 7 days (RV group) were compared to age-matched febrile infants without RVs (control group). Each group had 95 infants with similar demographics and medical history. RVs were primarily due to positive cultures and persistent fever. The control group had higher initial hospitalization rates, longer PED stays, and increased antibiotic treatment. Prevalence of serious bacterial infections (SBIs) did not significantly differ. Higher hospitalization, prolonged PED stays, and initial antibiotic treatment were associated with reduced RV incidence despite similar SBI rates. Return visits in infants <90 days were primarily driven by persistent fever and positive cultures. Addressing these factors through targeted parental education and improved care protocols may reduce RVs.

This chapter develops a puzzle about one of the central tenets of Judaism: that God revealed the Torah at Mount Sinai. The puzzle arises from a combination of more specific views, each of which is widely held in the Jewish theological tradition. First, that the realization of the Torah is the purpose for which the world was created. Second, that we couldn’t on our own discover the truths in which the Torah consists. Third and finally, that the Torah is a sort of wisdom; indeed, that once revealed we can see the wisdom in it. The combination of these views is puzzling. If the Torah is a kind of wisdom, it seems that God could have endowed us with cognitive faculties powerful enough to discover it on our own. Given that the Torah is the purpose of creation, it seems that God would have done so. But God didn’t. It looks like creation wasn’t properly matched to its purpose. The author of this chapter suggests two ways of solving the puzzle, both of which contend that while a way of life in keeping with the Torah exhibits great wisdom, it’s not the kind of thing that a wise individual could discover. One, drawing on the work of Hasdai Crescas, points to the demands of love, and love of God in particular. The other, drawing on the work of Nahmanides, points to the intrinsic systematicity of philosophy and theology, and the collective nature of the Sinaitic revelation.

Vegetation productivity is a critical indicator of global ecosystem health and is impacted by human activities and climate change. A wide range of optical sensing platforms, from ground-based to airborne and satellite, provide spatially continuous information on terrestrial vegetation status and functioning. As optical Earth observation (EO) data are usually routinely acquired, vegetation can be monitored repeatedly over time, reflecting seasonal vegetation patterns and trends in vegetation productivity metrics. Such metrics include gross primary productivity, net primary productivity, biomass, or yield. To summarize current knowledge, in this paper we systematically reviewed time series (TS) literature for assessing state-of-the-art vegetation productivity monitoring approaches for different ecosystems based on optical remote sensing (RS) data. As the integration of solar-induced fluorescence (SIF) data in vegetation productivity processing chains has emerged as a promising source, we also include this relatively recent sensor modality. We define three methodological categories to derive productivity metrics from remotely sensed TS of vegetation indices or quantitative traits: (i) trend analysis and anomaly detection, (ii) land surface phenology, and (iii) integration and assimilation of TS-derived metrics into statistical and process-based dynamic vegetation models (DVMs). Although the majority of used TS data streams originate from data acquired from satellite platforms, TS data from aircraft and unoccupied aerial vehicles have found their way into productivity monitoring studies. To facilitate processing, we provide a list of common toolboxes for inferring productivity metrics and information from TS data. We further discuss validation strategies of the RS data derived productivity metrics: (1) using in situ measured data, such as yield; (2) sensor networks of distinct sensors, including spectroradiometers, flux towers, or phenological cameras; and (3) inter-comparison of different productivity metrics. Finally, we address current challenges and propose a conceptual framework for productivity metrics derivation, including fully integrated DVMs and radiative transfer models here labelled as “Digital Twin”. This novel framework meets the requirements of multiple ecosystems and enables both an improved understanding of vegetation temporal dynamics in response to climate and environmental drivers and enhances the accuracy of vegetation productivity monitoring.

Specific phosphorylation patterns control the activity of multiphosphorylated proteins. In case of the Tau protein, multiphosphorylation leads to the formation of different disease-related condensates and aggregates. Studying the role of these specific patterns at the protein level is crucial for understanding the molecular mechanisms of Tauopathies such as Alzheimers Disease. However, due to the extreme difficulty in obtaining recombinant proteins with specific phosphorylation patterns using kinase-based methods, it is practically impossible to study the connection between specific phosphorylation patterns and aggregation events at the protein level. Here we addressed this problem by reducing the system to the peptide level and studying the effect of specific phosphorylation patterns on the condensation and aggregation of a specific domain of Tau, R4 (residues 336-358). To achieve this aim, we have applied advanced methods to synthesize a library of multiphosphorylated peptides derived from R4. We showed that specific phosphorylation patterns stringently control the formation of Tau aggregates and condensates. Phosphorylation of Ser341 promoted aggregation of R4 while phosphorylation of Ser352 promoted its condensation. Interestingly, Ser356 phosphorylation inhibited both processes, which can be overridden by double-phosphorylation at Ser341/Ser352. Differences between the microenvironments of the phosphorylated residues lead to their different effects on R4 aggregation upon phosphorylation. Our results show that working at the domain level using advanced peptide synthesis methods is a highly useful and practical way to provide valuable information about the effects of post translational modifications on protein activity.

The Sahara is the largest and driest of the hot deserts on Earth, with regions where rainfall reaches the surface on average less than once a year. Water resources are scarce, and rainfall tends to occur sporadically in space and time. While rain is a precious resource in the Sahara, heavy precipitation events (HPEs) in the desert have the potential to trigger flash floods on the barren soil. Because of the sparse rainfall monitoring network and the relatively poor performance of global models in representing rainfall over the Sahara, the analysis of Saharan HPEs has primarily relied on case studies. Therefore, general rainfall characteristics of Saharan HPEs are unexplored, and the prevailing weather conditions enabling such rainfall are unknown. To tackle this problem, we utilised satellite-derived precipitation estimations (IMERG) spanning 21 years (2000–2021) to identify ∼42⋅103 small (>103km2) to large (<106km2) HPEs in the Sahara and to extract their rainfall properties, and atmospheric reanalyses (ERA5) to examine the corresponding meteorological conditions in which they develop. Three case studies illustrate the relevance of cyclones for exceptionally large HPEs, including one in the driest region of the Sahara. Saharan HPEs occur, on average, every second day. They are more common in summer than in the other seasons, occur most frequently in the southern Sahara, and exhibit a clear convectively-driven diurnal cycle. Winter events have, on average, larger spatial extent, longer duration, and are characterised by larger areas exhibiting more extreme rainfall in terms of return periods. Autumn HPEs are concentrated in the western Sahara, while events in the north of the desert and in its driest core in the northeast occur mainly in winter and spring. In these regions, north of the Tropic of Cancer, events are highly associated with surface cyclones. HPEs that were associated with cyclones are characterised by larger spatial extent and rainfall volume. Considering that weather and climate models often depict synoptic-scale weather systems more accurately than rainfall patterns, the association of Saharan HPEs with surface cyclones and other synoptic-scale systems can aid in comprehending the effects of climate change in the desert. Furthermore, it underscores the potential for higher predictability of these events.
