Innovative approaches are urgently required to alleviate the growing pressure on agriculture to meet the rising demand for food. A key challenge for plant biology is to bridge the notable knowledge gap between our detailed understanding of model plants grown under laboratory conditions and the agriculturally important crops cultivated in fields or production facilities. This Perspective highlights the recent development of new analytical tools that are rapid and non-destructive and provide tissue-, cell- or organelle-specific information on living plants in real time, with the potential to extend across multiple species in field applications. We evaluate the utility of engineered plant nanosensors and portable Raman spectroscopy to detect biotic and abiotic stresses, monitor plant hormonal signalling as well as characterize the soil, phytobiome and crop health in a non- or minimally invasive manner. We propose leveraging these tools to bridge the aforementioned fundamental gap with new synthesis and integration of expertise from plant biology, engineering and data science. Lastly, we assess the economic potential and discuss implementation strategies that will ensure the acceptance and successful integration of these modern tools in future farming practices in traditional as well as urban agriculture.

The properties of clays and oxides govern many environmental processes, consequently, ongoing effort is invested in developing non-destructive, in-situ analytical tools that reflect these properties. Herein, the physicochemical properties of montmorillonite (MMT) and iron-oxide coated montmorillonite (FeOx-MMT) were characterized using common analytical techniques, and the results were compared to spectral induced polarization (SIP) measurements. FeOx-MMT particles showed a lower CEC, higher pH dependency of the surface charge, and lower suspension stability. Also, the size of the primary particles increased following iron-oxide deposition. SIP measurements over a range of salinities showed that the effective polarization length of the clays was in the order of several microns, suggesting the measurements of aggregates (not primary particles). Moreover, FeOx-MMT particles were more compact than MMT, and their size decreased with increasing salinity due to compaction of the EDL and arrangement of primary particles in the aggregate. The SIP-response to pH changes agreed with zeta potential measurements; at low pH values, MMT exhibited higher polarization due to the higher CEC. However, at a high pH, the differences diminish due to deprotonation of the Fe-OH surface groups. These findings suggest that SIP is a sensitive method that can detect changes in the surface chemistry of soil particles. (C) 2020 Elsevier Inc. All rights reserved.

SPX(1) has been identified in each class of vertebrates, and a second SPX (named SPX2) has been found in some non-mammalian species. We have cloned two spexin paralogs, designated as Spx1a and Spx1b, from Nile tilapia () that have varying tissue distribution patterns. Spx1b is a novel peptide only identified in cichlid fish, and is more closely related to Spx1 than Spx2 homologs as supported by phylogenetic, synteny, and functional analyses. Kisspeptin, Spx, and galanin (Gal) peptides and their corresponding kiss receptors and Gal receptors (Galrs), respectively, are evolutionarily related. Cloning of six tilapia Galrs (Galr1a, Galr1b, Galr2a, Galr2b, Galr type 1, and Galr type 2) and subsequent second-messenger reporter assays for Gα, Gα, and Gα suggests that Gal and Spx activate Galr1a/Galr2a and Galr2b, respectively. A decrease in plasma follicle stimulating hormone and luteinizing hormone concentrations was observed with injections of Spx1a or Spx1b . Additionally, application of Spx1a and Spx1b to pituitary slices decreased the firing rate of LH cells, suggesting that the peptides can act directly at the level of the pituitary. These data collectively suggest an inhibitory mechanism of action against the secretion of gonadotropins for a traditional and a novel spexin paralog in cichlid species.

Two covalently linked donor–acceptor copper phenanthroline complexes (C–A dyads) of interest for solar energy conversion/storage schemes, [Cu(i)(Rphen(OMV)24+)2]9+ = RC+A48+ with RC+ = [Cu(i)Rphen2]+ involving 2,9-methyl (R = Me) or 2,9-phenyl (R = Ph)-phenanthroline ligands that are 5,6-disubstituted by 4-(n-butoxy) linked methylviologen electron acceptor groups (A2+ = OMV2+), have been synthesized and investigated via quantum chemical calculations and nanosecond laser flash spectroscopy in 1,2-difluorobenzene/methanol (dfb/MeOH) mixtures. Upon photoexcitation, charge transfer (CT) states RC2+A+A36+ are formed in less than one ns and decay by charge recombination on a time scale of 6–45 ns. The CT lifetime of RC2+A+A36+ has a strong dependence on MeOH solvent fraction when R = Me, but is unaffected if R = Ph. This solvent effect is due to coordination of MeOH solvent in MeC+A48+ (i.e. exciplex formation) allowed by conformational flattening of the ligand sphere, which cannot occur in PhC+A48+ having bulkier Phphen ligand framework. Interestingly, the decay time of the CT state increases for both species at low magnetic fields with a maximum increase of ca. 30% at ca. 150 mT, then decreases as the field is increased up to 1500 mT, the highest field investigated. This magnetic field effect (MFE) is due to magnetic modulation of the spin dynamics interconverting 3CT and 1CT states. A quantitative modeling according to the radical pair mechanism involving ab initio multireference calculations of the complexes revealed that the spin process is dominated by the effect of Cu hyperfine coupling. The external magnetic field suppresses the hyperfine coupling induced spin state mixing thereby lengthening the CT decay time. This effect is counteracted by the field dependent processes of T0–S mixing through the Δg-mechanism and by a local mode spin–orbit mechanism. Further, the maximum MFE is limited by a finite rate of direct recombination of 3CT states and the spin-rotational mechanism of spin relaxation. This study provides a first comprehensive characterization of Cu(ii)-complex spin chemistry and highlights how spin chemistry can be used to manipulate solar energy harvesting and storage materials.

Breast cancer is the second leading cause of death in women above 60 years in the US. Screening mammography is recommended for women above 50 years; however, 22% of breast cancer cases are diagnosed in women below this age. We set out to develop a test based on the detection of cell-free RNA from saliva. To this end, we sequenced RNA from a pool of ten women. The 1254 transcripts identified were enriched for genes with an annotation of alternative pre-mRNA splicing. Pre-mRNA splicing is a tightly regulated process and its misregulation in cancer cells promotes the formation of cancer-driving isoforms. For these reasons, we chose to focus on splicing factors as biomarkers for the early detection of breast cancer. We found that the level of the splicing factors is unique to each woman and consistent in the same woman at different time points. Next, we extracted RNA from 36 healthy subjects and 31 breast cancer patients. Recording the mRNA level of seven splicing factors in these samples demonstrated that the combination of all these factors is different in the two groups (p value = 0.005). Our results demonstrate a differential abundance of splicing factor mRNA in the saliva of breast cancer patients.

The Soil Continuum Model questions the occurrence of any independent natural process of secondary synthesis that generates compounds structurally distinct from plant or microbial metabolites. This review shows that a vast volume of interdisciplinary scientific evidence supports the formation of relevant non-pre-existing complex molecules exhibiting various types of structures. These molecules form during degradation and decay of biological cell components. The spontaneous abiotic and enzymatically catalysed reactions of components of organic residues and of their oxidative decomposition products suggested by state-of-the-art studies are indeed those proposed by most of the classical descriptions of humification. The review also highlights the chemically active role of pedofauna, explaining why the apparently harsh conditions of alkaline extraction of HS cannot be considered un-natural. Many insects and larvae feeding on foliage of plants with a high content of tannins have a midgut pH above 9. Albeit, reducing conditions are often maintained to avoid oxidation, peroxidases are active in the intestinal tract and pass on to feces. Polyphenols are then immediately enzymatically oxidized to their reactive quinone form, once feces are excreted and exposed to oxygen. Implications of our current knowledge on the reactivity of plant components in soil are discussed in relation with the present state of the art research on humic substances. Contrary to claims by the Soil Continuum Model theory, complimentary modern approaches need to be used to understand the complexity of soil organic matter.

Scanning nanoscale superconducting quantum interference devices (nanoSQUIDs) are of growing interest for highly sensitive quantitative imaging of magnetic, spintronic, and transport properties of low-dimensional systems. Utilizing specifically designed grooved quartz capillaries pulled into a sharp pipette, we have fabricated the smallest SQUID-on-tip (SOT) devices with effective diameters down to 39 nm. Integration of a resistive shunt in close proximity to the pipette apex combined with selfaligned deposition of In and Sn, have resulted in SOT with a flux noise of 42 n0Hz–1/2, yielding a record low spin noise of 0.29 µBHz–1/2. In addition, the new SOTs function at sub-Kelvin temperatures and in high magnetic fields of over 2.5 T. Integrating the SOTs into a scanning probe microscope allowed us to image the stray field of a single Fe3O4 nanocube at 300 mK. Our results show that the easy magnetization axis direction undergoes a transition from the (111) direction at room temperature to an in-plane orientation, which could be attributed to the Verwey phase transition in Fe3O4.

ABSTRACTSocial media constitute a fertile though challenging arena for the use of ironic humor. A combination of facilitating and hindering factors turns the production and identification of irony in this sphere into a complex venture, positioning it as a powerful tool in consolidating group boundaries. The main aims of this paper are to identify the markers of ironic humor on social media and to explicate their workings within the dynamics of digital interactions. Existing literature about ironic markers addresses mostly face-to-face and mass-mediated interactions, while there is a gap in our understanding of the production and interpretation of irony in the unique communicative conditions of social media. An analysis of a bilingual corpus of successful and failed ironic utterances extracted from five social network sites yielded a novel typology of five ironic markers: platform, participants, style, intra-textual content, and contextual knowledge. These markers both resemble and deviate from features of irony in non-digital settings. Media affordances often allow access to necessary complementary information, yet such forensic activity is only accessible to active users, who recognize the need for further investigation of the meant. Hence, using certain types of ironic markers forms utterances that are decipherable to some audiences but opaque to others. In this sense, the marking of digital irony and its decoding both rely on group boundaries and play a central role in their delineation. We conclude by evaluating the roles of this new regime of ironic markers in processes of boundary work.

BACKGROUND: Bleeding disorders (BD) are under-recognized in adolescents with heavy menstrual bleeding (HMB). OBJECTIVES: The lack of clinical guidelines and variable symptomatic management of HMB created the imperative to standardize HMB care to identify and manage BD in adolescents. METHODS: We convened an international working group (WG), utilized the results of a literature review to define knowledge gaps in HMB care, and used the collective clinical experience of the WG to develop care considerations for adolescents with BD and HMB. We then solicited input on the appropriateness of HMB care considerations from expert stakeholders representing hematology, adolescent medicine, and obstetrics-gynecology. We conducted an expert panel online, using the ExpertLens platform. During a three-round online modified-Delphi process, the expert panel rated the appropriateness of 21 care considerations using a 9-point scale to designate care as appropriate (7-9), uncertain (4-6), or inappropriate (1-3) covering screening for BD, the laboratory work-up, and management of adolescents with BD that present with HMB. We used the RAND/UCLA appropriateness method to determine the existence of consensus among the interdisciplinary panel of experts. RESULTS: Thirty-nine experts participated in the panel. The experts rated fifteen HMB care considerations as appropriate, six as uncertain, and none as inappropriate. CONCLUSIONS: The HMB care statements represent the first set of HMB care considerations in adolescents with BD, developed with broad expert input on appropriateness. Although likely to be of interest to a range of clinicians who routinely manage adolescents with HMB, additional research is required in many key areas.

Contemporary media production in East Asia has been booming in recent decades, reaching consumers of various nationalities to an extent unseen before. Concurrently, various collaborations in the production and promotion of movies, TV programs, animation, videogames, music, and the like have intensified and become more effective in delivering cultural content across national borders. The state in East Asia, once considered a custodian of local culture and a censor of foreign influences, has gradually changed its position and now views media production and its export as an integral part of market activity beneficial for both economy and diplomacy. Drawing upon governmental reports and interviews with government officials and media industry personnel in Korea and Japan, this paper examines the way the state reacts to the massive production and export of media culture, its attempt to integrate it within its developmental economic model, and the bureaucratic restructuring this process entails. The central argument presented is that state policy toward the media sector, and the abundance of institutions dealing with this sector in Korea and Japan, are the result of the growth of market-led media production and globalization. In other words, media industry development is not the result of policy, policy is the result of media industry development. In a wider context, this paper aims to better conceptualize the relations between state policy and media production in East Asia and the dynamics of industrial globalization under state guidance.

Non-ribosomal peptides (NRPs) and polyketides (PKs) are among the most profuse families of secondary metabolites (SM) produced by bacteria. These compounds are believed to play an important ecological role in microbe-microbe and microbe-plant interactions in soil and roots microbiomes. Over the years, screening of NRPs and PKs in soil bacteria has resulted in high rates of rediscovery, mainly due to challenges associated with bacterial isolation. The quest to expose compounds in the hidden ``unculturable'' fraction of the soil microbiome, and to activate existing and novel SM gene clusters in cultivated bacteria, has facilitated a paradigm shift from traditional isolation-based natural product identification platforms to novel `ecologically inspired' culturing techniques and cutting-edge culture-independent methods. This review provides a comprehensive overview of platforms and applications for studying secondary metabolites in soil and root environments, deliberating the benefits and limitations of the various approaches. Initially, it highlights innovative methodologies to ``culture the unculturable'' to uncover novel soil bacterial SM. Next, it explores ``culture-independent'' platforms for the identification of SM-synthesizing gene clusters through next generation sequencing and bioinformatics. It then evaluates innovative approaches for heterologous expression of metabolites from complex soil environments. Finally, it presents a conceptual integrated pipeline for evaluating the potential function and role of root-associated bacterial SM in suppressive soils that inhibit plant pathogens. This pipeline can be modified to address the ecological role of SM in other soil and root ecosystems, which can ultimately enhance our understanding of microbe-microbe and bacterial-plant interactions.

Acute and chronic physical exercises may enhance the development of statin-related myopathy. In this context, the recent (2019) guidelines of the European Society of Cardiology (ESC) and the European Atherosclerosis Society (EAS) for the management of dyslipidemias recommend that, although individuals with dyslipidemia should be advised to engage in regular moderate physical exercise (for at least 30 min daily), physicians should be alerted with regard to myopathy and creatine kinase (CK) elevation in statin-treated sport athletes. However it is worth emphasizing that abovementioned guidelines, previous and recent ESC/EAS consensus papers on adverse effects of statin therapy as well as other previous attempts on this issue, including the ones from the International Lipid Expert Panel (ILEP), give only general recommendations on how to manage patients requiring statin therapy on regular exercises. Therefore, these guidelines in the form of the Position Paper are the first such an attempt to summary existing, often scarce knowledge, and to present this important issue in the form of step-by-step practical recommendations. It is critically important as we might observe more and more individuals on regular exercises/athletes requiring statin therapy due to their cardiovascular risk.
