The reconnection of two interacting vortex tubes is a fundamental process in fluid mechanics, which, at very high Reynolds numbers, is associated with the formation of intense velocity gradients. Reconnection is usually studied using two antiparallel tubes which are destabilized via the long-wavelength Crow instability, leading to a very symmetric configuration and to a strong flattening of the cores into thin sheets. Here, we consider the interaction of two initially straight tubes at an angle of β and show that by relaxing some symmetries of the problem, a rich phenomenology appears. When the angle between the two tubes is close to β=90∘, their interaction leads to pairing of small portions of antiparallel tubes, followed by the formation of thin and localized vortex sheets as a precursor of reconnection. The subsequent breakdown of these sheets involves a twisting of the paired sheets, followed by the appearance of a localized cloud of small-scale vortex structures. By decreasing β, we show that reconnection involves increasingly larger portions of tubes, whose cores are subsequently destabilizing, leaving behind more small-scale vortices. At the smallest values of the angle β studied, the two vortices break down through a mechanism, which leads to a cascadelike process of energy conveyance across length scales similar to what was found for previous studies of antiparallel vortex tubes (β=0) which imposed no symmetries. While, in all cases, the interaction of two vortices depends on the initial condition, the rapid formation of fine-scale vortex structures appears to be a robust feature, possibly universal at very high Reynolds numbers.

A proliferation inducing ligand (APRIL) mediates a key role in the generation and survival of antibody-inducing plasmocytes. Based on this, APRIL has been targeted in autoimmune diseases including multiple sclerosis (MS) and optic neuritis (ON). In MS lesions, APRIL has a new cellular target, the reactive astrocyte and mediates an immunosuppressive activity. Here, we analyzed APRIL expression in a case of neuromyelitis optica (NMO), another autoimmune neurodegenerative disease, showing selective aquaporin-4 depletion in the spinal cord, complement deposition and infiltration of polymorphonuclear cells. We analyzed by immunohistochemistry the presence of APRIL-producing cells, plasmocytes, astrocytes and the localization of secreted APRIL in a lesion from NMO. Plasmocytes were present close to APRIL-producing cells in meninges. However, our main observation was that APRIL targets reactive astrocytes in this lesion of NMO similarly to MS.

Causal Selection is a widely discussed topic in philosophy and the cognitive sciences, concerned with characterizing the choice of “the cause” among the many individually necessary and jointly sufficient conditions on which any effect depends on. In this paper, we argue for an additional selection process underlying causal state-ments: Causative-Construction Selection, which pertains to the choice of linguistic constructions used to express causal relations. By exploring this phenomenon, we aim to answer the following question: given that a speaker wishes to describe the relation between one of the conditions and the effect, which linguistic constructions are avail-able? We take CC-selection to be more crucial than causal selection, since the latter is in fact restricted by the linguistic options resulting from the former. Based on a series of experiments, we demonstrate that factors taken previously as contributing to causal selection should, in fact, be considered as the parameters that license the various lin-guistic constructions under given circumstances, based on previous knowledge about the causal structure of the world (the causal model). These factors are therefore part of the meaning of the causative expressions.

The osteological study of eight well-dated horse skeletons from Xinjiang (350 BCE) is reported as an important discovery: the earliest direct evidence for horseback riding in China. However, the sites are located roughly 2,000 km from the centers of Chinese civilization, where evidence for domesticated horses predates these skeletons by some 1,000 years, and artistic depictions of horseback riding appear some 400 years earlier than these skeletons. Horses in China were extensively used for charioteering: The duke of Qi’s tomb complex in Shandong (490 BCE) included several hundred sacrificial horses. Why, then, is this discovery so important?

Its importance, we argue, lies elsewhere: the beginning of cavalry warfare, a phenomenon that changed the geopolitical makeup of this region for the next two millennia. The occurrence rate of vertebral abnormalities suggests extensive exploitation, which is commonly attributed to excessive horseback riding, and their dating correlates with the onset of cavalry warfare on China’s northern frontiers.

Gout is an inflammatory disease triggered by deposition of monosodium urate (MSU) crystals in the joints, resulting in high neutrophil influx and pain. Here, we studied the role of the inhibitory receptor CD300a in the resolution process in a murine model of gout. We found increased CD300a expression on neutrophils emigrated to the joint. When compared to WT mice, CD300a−/− mice had persistent neutrophil influx till 24 hr after MSU injection. This was associated with increased concentration of IL-1$\beta$ and greater tissue damage in the joints of CD300a−/− mice. There was an increase in the percentage of apoptotic neutrophils in the synovial lavage of WT mice, as compared to CD300a−/− mice. This difference was reflected in the decline of efferocytic events in the synovial cavity of CD300a−/− mice 24 hr after MSU injection. A CD300a agonistic antibody was shown, for the first time, to increase apoptosis of human neutrophils, and this was associated with cleavage of caspase-8. Cellulose nanocrystals (CNC) are well-suited to the preparation of biocomposite films and packaging material due to its abundance, renewability, biodegradability, and favorable film-forming capacity. In this study, different CNC and corn zein (CZ) composite films were prepared by adding CZ to the CNC suspension prior to drying, in order to change internal structure of resulting films. Films were developed to examine their performance as an alternative water vapor and oxygen-barrier for flexible packaging industry. Water vapor permeability (WVP) and oxygen transmission rate (OTR) of the biocomposite films decreased significantly in a specific ratio between CNC and CZ combined with 1,2,3,4-butane tetracarboxylic acid (BTCA), a nontoxic cross linker. In addition to the improved barrier properties, the incorporation of CZ benefitted the flexibility and thermal stability of the CNC/CZ composite films. The toughness increased by 358%, and Young’s modulus decreased by 32% compared with the pristine CNC film. The maximum degradation temperature increased by 26 °C, compared with that of CNC film. These results can be attributed to the incorporation of a hydrophobic protein into the matrix creating hydrophobic interactions among the biocomposite components. SEM and AFM analysis indicated that CZ could significantly affect the CNC arrangement, and the film surface topography, due to the mechanical bundling and physical adsorption effect of CZ to CNC. The presented results indicate that CNC/CZ biocomposite films may find applications in packaging, and in multi-functionalization materials.

The yolk sac (YS) consists of the yolk, which supplies nutrients, and the YS tissue, which surrounds the yolk and provides essential metabolic functions for the developing embryo. The YS tissue is derived from the midgut of the embryo and consists of a layer of endodermal epithelial cells (EEC) in contact with the yolk contents, a mesodermal layer that contains the vascular system and an outer ectodermal layer. The YS tissue is a multifunctional organ that provides essential functions such as host immunity, nutrient uptake, carbohydrate and lipid metabolism, and erythropoiesis. The YS tissue plays a role in immunity by the transport of maternal antibodies in the yolk to the embryonic circulation that feeds the developing embryo. In addition, the YS tissue expresses high mRNA levels of the host defense peptide, avian beta-defensin 10 during mid embryogenesis. Owing to its origin, the YS EEC share some functional properties with intestinal epithelial cells such as expression of transporters for amino acids, peptides, monosaccharides, fatty acids, and minerals. The YS tissue stores glycogen and expresses enzymes for glycogen synthesis and breakdown and glucogenesis. This carbohydrate metabolism may play an important role in the hatching process. The mesodermal layer of the YS tissue is the site for erythropoiesis and provides erythrocytes before the maturation of the bone marrow. Other functions of the YS tissue involve synthesis of plasma proteins, lipid transport and cholesterol metabolism, and synthesis of thyroxine. Thus, the YS is an essential organ for the growth, development, and health of the developing embryo. This review will provide an overview of the studies that have investigated the functionalities of the YS tissue at the cellular and molecular levels with a focus on chickens.

Simple Summary:& nbsp;This study presents in silico models of neurokinin B receptor tiTac3Ra (tilapia Tachykinin 3 receptor a) and its potential binding sites, as well as docking native tilapia Neurokinin F (tiNKF) and tilapia Neurokinin B (tiNKB) to theses orthosteric binding sites. For a better understanding of the binding confirmation and interaction of the structures, we compared the conformation between peptide docking and induced fit docking results. We have tried to analyze the affinity of binding and binding site interactions parallel to in-vitro results of receptor activity. NKB antagonists inhibit male tilapia gonadal development and gonadotropin release. We further verified the in-vivo effect of the antagonists on gonadal development in males. Studying the receptor activity of variants with alanine mutations at Phe251(6.44) and Met289(7.43) respectively, we found that, while both variants completely underperformed, there was no direct interaction with the ligand in the binding process indicating their role in post binding receptor activation rather than the binding process itself.
NKB (Neurokinin B) is already known to play a crucial role in fish reproduction, but little is known about the structure and function of NKB receptors. Based on an in silico model of the tilapia NKB receptor Tachykinin 3 receptor a (tiTac3Ra) found in the current study, we determined the key residues involved in binding to tilapia NKB and its functional homologue NKF (Neurokinin F). Despite studies in humans suggesting the crucial role of F251(6.44) and M289(7.43) in NKB binding, no direct peptide interaction was observed in tilapia homologs. In-silico, Ala mutations on residues F251(6.44) and M289(7.43) did not influence binding affinity, but significantly affected the stability of tiTac3Ra. Moreover, in vitro studies indicated them to be critical to tiNKB/tiNKF-induced receptor activity. The binding of NKB antagonists to tiTac3Ra both in-vitro and in vivo inhibits FSH (follicle stimulating hormone) and LH (luteinizing hormone) release and sperm production in mature tilapia males. Non-peptide NKB antagonist SB-222200 had a strong inhibitory effect on the Tac3Ra activation. SB-222200 also decreased LH plasma levels; two hours post intraperitoneal injection, changed sperm volume and the ratios of the different stages along the spermatogenesis in tilapia testes.
